Tanshinone, also known as total tanshinone, is a lipid soluble phenanthraquinone compound with bacteriostatic effect extracted from traditional Chinese medicine Salvia miltiorrhiza (Salvia miltiorrhiza root), from which more than 10 tanshinone monomers such as tanshinone I, tanshinone IIA, tanshinone IIB, cryptotanshinone and ISO cryptotanshinone are separated, including cryptotanshinone, dihydrotanshinone II, hydroxytanshinone, methyl tanshinate The five monomers of tanshinone Ⅱ B have antibacterial effect, anti-inflammatory and cooling effect. Sodium tanshinone IIA sulfonate, the sulfonated product of tanshinone IIA, can be dissolved in water. The clinical trial shows that it has significant effect in the treatment of angina pectoris and little side effects. It is a new drug for the treatment of coronary heart disease. Tanshinone has many effects, such as antibacterial, anti-inflammatory, promoting blood circulation and removing blood stasis, promoting wound healing, etc. no obvious side effects have been observed after long-term use.
Pharmacological Action
1. Antibacterial effect in vitro, tanshinone has stronger antibacterial effect on Staphylococcus aureus than berberine, and also has inhibitory effect on Mycobacterium tuberculosis H37Rv strain (the minimum inhibitory concentration is less than 1.5mg/ml) and two kinds of Trichophyton.
2. Anti inflammatory effect tanshinone has obvious anti-inflammatory effect in rats. In the first stage of inflammation model, it can significantly inhibit the increase of capillary permeability caused by histamine; It has inhibitory effect on acute joint swelling caused by egg white, carrageenan and dextran; It has inhibitory effect on exudative formaldehyde peritonitis. In the second stage of inflammation model, it can significantly inhibit the leukocyte migration induced by gelatin in mice; It has obvious inhibitory effect on subacute formaldehyde arthritis. Cotton ball granuloma test had no inhibitory effect on the third stage inflammation model (i.e. no effect in chronic stage). Tanshinone can increase PGF2 in blood of rats α And PGE levels decreased, which may be related to the anti-inflammatory mechanism of tanshinone.
3. Anticoagulant effect tanshinone has anticoagulant effect. The effect is stronger than that of protocatelaldehyde.
4. Other effects tanshinone can improve the survival rate or prolong the survival time of mice under low-pressure hypoxia. Tanshinone has anti androgen effect, reduces sebum production, has anti Propionibacterium acne effect, and reduces inflammation exacerbated by bacterial metabolism. It can be used to treat acne. Tanshinone has antipyretic effect on rabbits caused by typhoid vaccine. Mouse experiments show that tanshinone has mild estrogen activity through ovary.
5. In vivo, observe the situation that tanshinone enters the liver and is excreted from the bile after duodenal administration. The results show that a small amount of tanshinone can be measured in the bile about 1h after duodenal administration, and the excretion peak is about 3h after administration. The excretion rate of tanshinone with different structures in liver is different. In addition, it was also found that after extracting tanshinone from bile with chloroform, denatured protein was precipitated from the aqueous solution of bile, and its content was related to different administration groups.
